I am not a business person so bear with me please.
If the example purchase order form above https://www.jotform.com/72437801358964 is used, what happens after it is submitted? How does the payment get to the business owner. Does a copy of the order get get printed and sent\mailed in with the payment?

The purchase order payment tool allows you to create payment forms without payment vendor integration. In other words, no actual payments will be processed.
You will receive the order to your email with the products, quantity and the total price. And then you can proceed with your own payment procedures.
If this isn't what you are looking for, you can try another payment process like PayPal or Authorize.net

My Form as of now has 2 amt fields on them. One is for payment if a check
will be mailed in with the form after it is printed.
The other field is for payment via PayPal or credit card.
As of now it looks like the company I am creating this form for, does not
have a PayPal account. That being said, can I just delete the PayPal field
option, and leave the other amt field or must I create a "Purchase Order"
field instead?

It is up to your needs, if PayPal doesn't work for you, you can choose Purchase Order or any other payment processor that JotForm offers:

grayed out. How do I un-select PayPal and then select Purchase order for a
field that is already defined? (Perhaps the other field is already
designated as a Purchase Order, but how can I tell if it is?

You have integrated Purchase Order in your form.
You can see in the screenshot that Purchase Order is your active Payment Integration.
If you want to change it (which I think you don't), you have to delete it and the other options will become available.

The Purchase Order Integration differs from other payment methods because no payments will be processed on form submission. It has almost all the other features that other payment methods have, like SHIPPING options, creating products, coupon codes, etc.
You can send the Bank Account information along with a Reference number on the email that is sent when the submission is made. So, no invoice is generated by default.

The field that you are interested in (Amount Enclosed) is a Simple Text Field. You can use that as the Total that the user has to pay.
